Sir Richard is reprising an advert in which a woman eagerly awaits her arrival by train at a station where her boyfriend is waiting for her.
In the original, the entrepreneur made a cameo appearance as a refuse collector but in the new version - a "viral" ad on YouTube - he takes a starring role.
As Caprice steps out of the train to greet Sir Richard she is rebuffed in favour of a homely woman with "Virgin" daubed across her chest.
As the couple walk off hand in hand the final shot shows "Goes Like a Train" painted on the woman's back.
Sir Richard said: "When Virgin Trains asked if I would support their first ad campaign to celebrate our increased frequency and speed, wi-fi, uninterrupted mobile phone connection and the fact that weekends are now back on the west coast mainline, I jumped at the chance."
Last month, the tycoon wore fake tattoos to play a binman in a TV advert at Glasgow Central Station, which drew criticism for its portrayal of staff there.
