Around 12 parents are staging a sit-in at Wyndford Primary in Maryhill, Glasgow, in protest at the planned demolition of the building as part of wider spending cuts.
Glasgow City Council voted to close 11 schools and nine nurseries at a meeting in April despite months of heated protests.
Council staff arrived on Saturday to remove furniture and computers from the school and were prevented from driving out of the playground after campaigners lay across the school exit. City Building workers were forced to secure the vans and leave the school empty handed.
The women have pledged to remain inside the building indefinitely. It is understood the school's gas and water supply has now been turned off but campaigners still have access to electricity.
Members of the Glasgow Save Our Schools Campaign occupied the school at Easter and the neighbouring St Gregory's Primary, which has also closed.
Glasgow City Council say the closures are necessary to tackle falling school rolls and crumbling buildings.
A council spokesman said: "The former Wyndford Primary is now closed and is no longer in operation as a school."
Nikki Rathmill, 36, who is among the parents taking part in the sit-in, said: "We have been told that the council are going to move a family and recreation centre into St Gregory's.
"If it is good enough to be used as a recreation centre then why are they closing the school?"
