Hamilton stayed top of the Scottish Premiership on goal difference despite Dundee ending their nine-game unbeaten run in the league at windswept Dens Park.
Two second-half goals saw Paul Hartley's side become the first team to defeat Alex Neil's league leaders since the first day of the season when they went down at home to Inverness.
David Clarkson's 69th-minute header was his fourth goal in as many matches and four minutes later team-mate Greg Stewart made sure of the points with a well-taken goal to clinch a 2-0 win.
Hamilton selected the same side which had beaten Aberdeen 3-0 in their previous league outing. And it was the league leaders who began brightest and created the first chance within three minutes.
Anthony Andreu's ball forward beat the Dundee offside trap and Mickael Antoine-Curier unleashed a powerful right-foot shot from the edge of the penalty area, which Scott Bain did well to get down for and turn wide.
Dundee responded after 12 minutes when full-back Paul McGinn whipped over a decent cross which striker Stewart met, only to see his downward header go into the arms of Accies goalkeeper Michael McGovern.
Hamilton had another opportunity to threaten the Dundee goal. Dougie Imrie's pass forward released Andreu through and he had time to steady himself but shot wide of the target with his left foot.
The visitors were slightly on top and after 27 minutes they went close to taking the lead.
Ali Crawford's in-swinging free-kick caught out the home defence and allowed Michael Devlin to ghost in at the far post, but his header hit the side-netting much to the hosts' relief.
On the half-hour mark, Dundee forged forward as Gary Harkins fed Paul McGowan whose deep cross picked out Clarkson.
The former Motherwell striker attempted to cut the ball back into the six-yard box but was off balance and the chance was lost.
Dundee's Kevin McBride became the first player to be booked for a late foul on Imrie.
Shortly after, Imrie had a snap shot well saved by Bain as the Lanarkshire club attempted to gain the lead before the break.
Two minutes before half-time, Hamilton's Grant Gillespie was handed a yellow card for a foul on Paul McGowan.
The second half started with Antoine-Curier trying his luck with a 20-yard free-kick which Bain held comfortably.
At the other end, Dundee defender Gary Irvine had a free header from Stewart's corner after 55 minutes. Irvine had more space than he thought but saw his downward header go well wide of McGovern's left-hand post.
Dundee tried to become more of an attacking force by replacing midfielder McBride with forward Martin Boyle.
Hamilton then made a double substitution as Jason Scotland and Louis Longridge took over from Daniel Redmond and Antoine-Curier.
But it was Dundee who struck through Clarkson.
Harkins' curling corner caught the visiting defence napping and Clarkson rose unchallenged to power a header high past McGovern.
And after 73 minutes they went 2-0 ahead when Harkins headed the ball forward into the path of Stewart.
With only McGovern to beat, Stewart kept his cool from 12 yards to send a low left-footed finish beyond McGovern and make it back-to-back wins for the Tayside club.
